Sarcoma osteogenic-like Saos-2 cells on the silica nanofibres coated with nanodiamond

Nanotechnologies are widely used in bone tissue engineering. Nanodiamond materials have excellent biomechanical properties and can be modified to obtain biomimetic properties. The aim of the study was to prepare silica nano-fibres coated with nanodiamond and to characterise them in vitro in terms of their biocompatibility and bioactivity.
Saos-2 cells were cultured on SiOx materials covered with nanodiamond for 7 days. Materials were as follows: SiOx nano-fibres covered with nanodiamond by chemical vapour deposition (CVD) (H-termination), SiOx nano-fibres with nanodiamond after plasma O-termination, and SiOx nano-fibres without nanodiamond coating. As a control, glass coverslips were used. On the days 1, 4, and 7 after seeding, cell number and cell area on each of material was evaluated. The cells were also stained for proteins involved in osteoblasts adhesion, i.e. ?-actin (on the day 1) and vinculin (on the day 4), and in osteogenic differentiation, i.e. osteocalcin (on the day 7). On the day 7 cell metabolic activity (by resazurin assay) and ALP activity of the cells was measured as well. The concentration of vinculin and osteocalcin are being measured by enzymatic immunosorbent assay in cell homogenates prepared on day 7 after seeding.
The silica nanofibres coated with both H- and O-terminated nanodiamond supported the adhesion and proliferation of Saos-2 cells. These materials seem to be biocompatible and suitable for bone tissue engineering.
